Background
CXCR4, also known as CD184, is a chemokine receptor belonging to the GPCR family. It plays a crucial role in immune cell trafficking and hematopoiesis.
The interaction of CXCR4 with its ligand, CXCL12 (stromal cell-derived factor 1), is pivotal for the migration and homing of hematopoietic stem cells and leukocytes.
CXCR4 serves as a co-receptor for HIV entry into cells.
In oncology, CXCR4 is implicated in tumor growth, metastasis, and angiogenesis. Its expression is often up-regulated in haematological malignancies, and correlates with therapy resistance and poor prognosis.
Targeting CXCR4 is viewed as a promising therapeutic strategy in haematology-oncology indications. Understanding the CXCR4-CXCL12 axis is crucial in cell-based assays for developing novel therapeutic strategies targeting this pathway.

Product Details
Stable recombinant CHO cells at low and high confluency, cultured in CHO Growth Medium A (INS-ME-1039) as a 2D monolayer.
Expression of CXCR4 in CHO-CXCR4 cells (INS-SF-1004) analyzed by Flow Cytometry.
Primary antibody: CD184 (CXCR4)-PE labelled, (eBioscience, Cat.No.: 12-9999-41) was used at 1:100 dilution as per manufacturer’s recommendations.
Flow Cytometry was performed according to the protocol provided under the „Protocols“ tab.
Quantification: approx. 400 molecules per cell (Low) and approx. 9500 molecules per cell (Medium-High) analyzed by BD QuantiBrite quantification kit (cat.no. 340495) according to the protocol provided under the „Protocols“ tab.
Controls: CHO control – Parental Landing Pad cells, stably transfected with an empty vector and stained with the same primary antibody as the target-expressing cells.
We recommend the following reagents for the cell culture of this cell line:
Growth Medium:

Material:
- PBS/EDTA solution
- 2% FBS/FCS in PBS (FACS Buffer)
- primary antibody (concentration is indicated under „Target expression“ tab)
Protocol:
Wash Protocol: Add FACS Buffer, resuspend cells gently, then centrifuge at 300×g for 5min.
- Prepare detection reagents in FACS buffer using the concentrations indicated under Materials.
- Aspirate medium from cells.
- Add PBS/EDTA solution to the cells and incubate at room temperature or 37°C for 5-10min, or until the cells detach.
- Wash cells 1×.
- Add primary antibody in FACS buffer, resuspend cells gently.
- Incubate at ambient temperature for 20-30min.
- Wash cells 2×.
- Resuspend in 100-200µl FACS Buffer.
- Analyse cells using a flow cytometer.

Important: All instrument settings for fluorescence and compensation must be the same as the cellular assay settings.
- Reconstitute the BD Quantibrite beads using 0.5 mL of buffer, such as phosphate buffered saline (PBS) with sodium azide plus 0.5% bovine serum albumin (BSA), and vortex.
- Measure fluorescence of singlet beads using a flow cytometer and extract the geometric means of all four fluorescence peaks.
- Measure cells using the same instrument settings.
- Use the lot-specific values for the PE molecules per bead and linear regression to calculate the PE molecules per cell (approximation of target molecules per cell)
- For an even more meaningful result, contact the antibody provider for information on the number of fluorescent molecules per antibody.

Yes, for selected targets, we provide cell lines that exhibit varying expression levels. Detailed information regarding the availability of different expression levels can be found in both the ‚Product Summary‘ and ‚Product Details‘ sections.
Cell lines with available expression level variations are denoted by an abbreviation following the catalog number. For instance, ‚INS-SF-1020-MH‘ denotes a cell line with medium-high expression levels, whereas ‚INS-SF-1020-L‘ indicates a cell line with low expression of the same target.
Should you require a specific expression level not readily available in our standard product range, we also offer custom cell line generation services utilizing our advanced Landing Pad technology.
